    Desde a década de 1980 (Kocijancic, 1996; Rogers, 1996; Brizzi, 2000), muito se tem discutido sobre os limites e possibilidades do uso de softwares nas salas de aula. Entretanto, ainda que algumas pesquisas apontem o envolvimento de inúmeros fatores que determinam o sucesso do uso das TIC’s na escola poucos estudos são realizados com o intuito de compreender quais fatores impedem que os professores façam o uso otimizado de novas tecnologias no contexto escolar. Além disso, poucas pesquisas investem na compreensão de dimensões didático-pedagógicas que influenciam a inserção das TIC’S no contexto escolar. Assim, o objetivo deste trabalho é apresentar a análise de dados coletados em uma aplicação piloto de um curso de Física para o Ensino Médio, que faz uso de um software de realidade virtual (RV)

    Transgenic Overexpression of LARGE Induces alpha-Dystroglycan Hyperglycosylation in Skeletal and Cardiac Muscle

    Background: LARGE is one of seven putative or demonstrated glycosyltransferase enzymes defective in a common group of muscular dystrophies with reduced glycosylation of alpha-dystroglycan. Overexpression of LARGE induces hyperglycosylation of alpha-dystroglycan in both wild type and in cells from dystroglycanopathy patients, irrespective of their primary gene defect, restoring functional glycosylation. Viral delivery of LARGE to skeletal muscle in animal models of dystroglycanopathy has identical effects in vivo, suggesting that the restoration of functional glycosylation could have therapeutic applications in these disorders. Pharmacological strategies to upregulate Large expression are also being explored.Methodology/Principal Findings: In order to asses the safety and efficacy of long term LARGE over-expression in vivo, we have generated four mouse lines expressing a human LARGE transgene. On observation, LARGE transgenic mice were indistinguishable from the wild type littermates. Tissue analysis from young mice of all four lines showed a variable pattern of transgene expression: highest in skeletal and cardiac muscles, and lower in brain, kidney and liver. Transgene expression in striated muscles correlated with alpha-dystroglycan hyperglycosylation, as determined by immunoreactivity to antibody IIH6 and increased laminin binding on an overlay assay. Other components of the dystroglycan complex and extracellular matrix ligands were normally expressed, and general muscle histology was indistinguishable from wild type controls. Further detailed muscle physiological analysis demonstrated a loss of force in response to eccentric exercise in the older, but not in the younger mice, suggesting this deficit developed over time. However this remained a subclinical feature as no pathology was observed in older mice in any muscles including the diaphragm, which is sensitive to mechanical load-induced damage.Conclusions/Significance: This work shows that potential therapies in the dystroglycanopathies based on LARGE upregulation and alpha-dystroglycan hyperglycosylation in muscle should be safe

    Towards a synthesized critique of neoliberal biodiversity conservation

    During the last three decades, the arena of biodiversity conservation has largely aligned itself with the globally dominant political ideology of neoliberalism and associated governmentalities. Schemes such as payments for ecological services are promoted to reach the multiple ‘wins’ so desired: improved biodiversity conservation, economic development, (international) cooperation and poverty alleviation, amongst others. While critical scholarship with respect to understanding the linkages between neoliberalism, capitalism and the environment has a long tradition, a synthesized critique of neoliberal conservation - the ideology (and related practices) that the salvation of nature requires capitalist expansion - remains lacking. This paper aims to provide such a critique. We commence with the assertion that there has been a conflation between ‘economics’ and neoliberal ideology in conservation thinking and implementation. As a result, we argue, it becomes easier to distinguish the main problems that neoliberal win-win models pose for biodiversity conservation. These are framed around three points: the stimulation of contradictions; appropriation and misrepresentation and the disciplining of dissent.     Downregulation of genes with a function in axon outgrowth and synapse formation in motor neurones of the VEGF(delta/delta) mouse model of amyotrophic lateral sclerosis

    Background: Vascular endothelial growth factor (VEGF) is an endothelial cell mitogen that stimulates vasculogenesis. It has also been shown to act as a neurotrophic factor in vitro and in vivo. Deletion of the hypoxia response element of the promoter region of the gene encoding VEGF in mice causes a reduction in neural VEGF expression, and results in adult-onset motor neurone degeneration that resembles amyotrophic lateral sclerosis (ALS). Investigating the molecular pathways to neurodegeneration in the VEGF(delta/delta) mouse model of ALS may improve understanding of the mechanisms of motor neurone death in the human disease. Results: Microarray analysis was used to determine the transcriptional profile of laser captured spinal motor neurones of transgenic and wild-type littermates at 3 time points of disease. 324 genes were significantly differentially expressed in motor neurones of presymptomatic VEGF(delta/delta) mice, 382 at disease onset, and 689 at late stage disease. Massive transcriptional downregulation occurred with disease progression, associated with downregulation of genes involved in RNA processing at late stage disease. VEGF(delta/delta) mice showed reduction in expression, from symptom onset, of the cholesterol synthesis pathway, and genes involved in nervous system development, including axonogenesis, synapse formation, growth factor signalling pathways, cell adhesion and microtubule-based processes. These changes may reflect a reduced capacity of VEGF(delta/delta) mice for maintenance and remodelling of neuronal processes in the face of demands of neural plasticity. The findings are supported by the demonstration that in primary motor neurone cultures from VEGF(delta/delta) mice, axon outgrowth is significantly reduced compared to wild-type littermates. Conclusions: Downregulation of these genes involved in axon outgrowth and synapse formation in adult mice suggests a hitherto unrecognized role of VEGF in the maintenance of neuronal circuitry. Dysregulation of VEGF may lead to neurodegeneration through synaptic regression and dying-back axonopathy

    Seeing environmental injustices: the mechanics, devices and assumptions of environmental sustainability indices and indicators

    At the heart of any colonization project, and therefore any move to de-colonize, are ways of seeing nature and society, that then allow particular ways of governing each. This is plainly visible in a number of tools that exist to measure progress towards (or regress from) environmental sustainability. The tools use indices and indicators constructed mostly by environmental scientists and ecologists. As such, they are not neutral scientific instruments: they reflect the worldviews of their creators. These worldviews depend on three dimensions: the values they prioritize, the explanatory theories they use and the futures they envision. Through these means different tools produce conflicting notions of the sustainability of our economies and societies. In this article, we shed light onto the theoretical and epistemological assumptions that lie behind key international sustainability indices and indicators: the Environmental Performance Index,Domestic Material Consumption, Material Intensity, the Material Footprint, the Carbon Footprint, the Ecological Footprint and CO2 emissions (territorial). The variables included in these indices, the way they are measured, aggregated and weighted all imply a particular way of understanding the relationships between economy, society and environment. This divergence is most clearly visible in the fact that some indices are negatively correlated with each other. Where one index might plot growing environmental sustainability, another shows its decline. Our results highlight that those devices and the theories informing them are particularly interesting for way how colonialism is materialized. Some of these measurements hide the material roots of prosperity and the ecological (and economic) distributional conflicts exported to the poorer countries by the global North, and others show how its production and consumption levels are reliant upon a socio-ecological 'subsidy' imposed on Southern countries. These subsidies represent injustices that present a primafacie case for decolonizing indices and indicators of environmental governance

    Prenatal muscle development in a mouse model for the secondary dystroglycanopathies

    The defective glycosylation of α-dystroglycan is associated with a group of muscular dystrophies that are collectively referred to as the secondary dystroglycanopathies. Mutations in the gene encoding fukutin-related protein (FKRP) are one of the most common causes of secondary dystroglycanopathy in the UK and are associated with a wide spectrum of disease. Whilst central nervous system involvement has a prenatal onset, no studies have addressed prenatal muscle development in any of the mouse models for this group of diseases. In view of the pivotal role of α-dystroglycan in early basement membrane formation, we sought to determine if the muscle formation was altered in a mouse model of FKRP-related dystrophy
